In Painter, I use a 'variable chalk' brush and the 'Grainy water' blender to lay down the blocks of colour ( over the rough line drawing ). Then on a new layer, I use the 'scratchboard tool' brush to pull out sharper lines and scratchy edges and dabs. I can always go back and soften the sharp lines down with the blender brush again if I need to...but that's basically the process I use to give it the sketchy paintwork look that I try to aim for normally. By the time you pull back from the artwork to look at it though, it hopefully all holds together as a tight image...
